Vanderbilt-Ingram Cancer Center investigators have identified three new genetic "hotspots" linked to colorectal cancer. These variants, reported Dec. 23 in an Advanced Online Publication in Nature Genetics, provide new insight into the biology of colorectal cancer ? and could represent new therapeutic targets for the disease.
Colorectal cancer is one of the most commonly diagnosed cancers worldwide ? and rates are particularly high in the United States and other developed countries. Genetics plays an important role in both sporadic and familial (inherited) forms of the disease. However, only about 6 percent of colorectal cancer cases are explained by the rare genetic variants known to confer high risk of colorectal cancer (as seen in familial forms of the disease).
Previous studies on the genetic basis of colorectal cancer have pinpointed several additional variants, but most of the studies were conducted in European/Caucasian populations.
"Looking at different ethnic groups is important because the genetic structures can be different enough that variants identified in one population do not explain risk in other populations," said Wei Zheng, M.D., Ph.D., MPH, an Ingram Professor of Cancer Research and senior author on the study. "Because of the difference in genetic structures and underlying environment exposures, it might be easier to discover some risk variants in studies conducted in non-European populations."
In 2009, Zheng and colleagues in several Asian countries established the "Asia Colorectal Cancer Consortium" to search for novel genetic risk factors for the disease. The consortium included populations in China, Korea and Japan.
Using an approach known as a "genome-wide association study" (or GWAS), Zheng and colleagues began searching for common variants linked to disease risk.
From genomic data obtained from 2,098 colorectal cancer cases and 5,749 controls, the researchers identified 64 variants, or "single nucleotide polymorphisms" (SNPs), that were associated with colorectal cancer.
The investigators then replicated these findings in another set of samples, narrowing down the number of disease-associated variants to four. Three of those four variants were also associated with colorectal cancer risk in a larger European sample.
"The findings from this study are relevant to both Asian and European populations," said Zheng. "Interestingly, these three susceptibility loci were not discovered in previous studies conducted in European-ancestry populations."
This study highlights the importance of conducting genetic studies in non-European populations to fully uncover the genetic basis for common diseases, including colorectal cancer, Zheng noted.
While the specific functions of these newly identified susceptibility loci are not clear yet, several important genes are located in the regions near the risk variants discovered in this study. For example, one risk variant is located near CCND2, the gene encoding cyclin D2, a member of the cyclin family of proteins that regulate the cell cycle. Cyclins have been linked to cancer, but research on the CCND2 gene has been limited. Therefore, the current findings suggest the need for further research on the role of other cyclins and cyclin-dependent kinases in carcinogenesis.
"These new discoveries are very exciting," Zheng said. "They will certainly lead to future studies regarding the biology of these regions and the translational potential of these findings in cancer prevention and treatment."

ALLENTOWN, Penn./KANSAS CITY, Missouri (Reuters) - Gun enthusiasts thronged to shows around the country on Saturday to buy assault weapons they fear will soon be outlawed after a massacre of school children in Connecticut prompted calls for tighter controls on firearms.
Reuters reporters went to gun shows in Pennsylvania, Missouri and Texas, and found long lines to get in the door, crowds around the dealer booths, a rush to buy assault weapons even at higher prices and some dealers selling out.
The busiest table at the R.K. Gun & Knife show at an exposition center near the Kansas City, Missouri airport was offering assault weapons near the entrance.
West Plains, Missouri dealer Keith's Guns sold out of about 20 AR-15 style assault rifles in a little over an hour, owner Keith Gray said.
An AR-15 type assault weapon was among the guns authorities believe suspect Adam Lanza stole from his mother to use in the massacre of 20 school children and six adults at a Newtown, Connecticut elementary school on December 14.
The killing of innocent children at the school shocked the nation and prompted a number of politicians including President Barack Obama to call for a ban on assault weapons and ammunition clips that allow the rapid firing of multiple bullets.
Rather than tighten gun ownership restrictions, the powerful lobby for gun rights, the National Rifle Association, on Friday called for armed guards at every school.
D.R. Woody was one of those able to purchase an assault weapon at the Kansas City show on Saturday. He bought the gun for target practice because he is concerned they soon will be banned. "I didn't expect to find one. No gun stores have them," said Woody of the AR-15 type of gun.
RUSH TO BUY
The story was the same in Allentown, Pennsylvania, where Shirley Donley, a gun shop owner from nearby Quakertown, had an endless stream of customers. "Everybody wants assault weapons," she said, adding that she had sold more than 100 of that type of gun since the Connecticut tragedy. "I'm sold out."
Assault weapon is a broad term commonly used to refer to semi-automatic or automatic weapons that can fire multiple bullets rapidly. From 1994 to 2004 certain assault weapons and ammunition clips of more than 10 bullets were illegal.
The ban was allowed to expire when Republican George W. Bush was in the White House.
Prices for assault weapons have surged since the Connecticut shooting. At the Kansas City show, Jerome Ratliff bought an AR-15 on Saturday for target practice, paying $925. The same model would have cost only about $400 a year ago, he said. Most models were selling for $1,500 or more.
Bob Hofmeister, whose wife owns Xtreme Sports, a gun dealer with a table at the Kansas city show, said the business sold 15 to 20 AR-15s in the past week.
"Some of these people just want to show their rights to own guns," Hofmeister said.
The Second Amendment to the U.S. Constitution guarantees the right to bear arms, and most gun enthusiasts at the shows on Saturday said more restrictions on guns would not stop mass killings such as Connecticut.
Adam Ouart of Mansfield, Texas stood in a line with about another dozen people at the Lone Star show in Forth Worth, Texas, in hopes of buying a gun.
"The answer is not to limit people having guns. If someone wants to hurt somebody they are going to find a way to do it," Ouart said.
Several dealers and buyers interviewed at the shows supported the NRA proposal to put armed guards in schools.
More than 200 people lined up at each of three entrances on Saturday morning to pay the $8 entrance fee to the Will Rogers Memorial Center in Fort Worth, which has an exhibit hall spanning 25 acres. They crowded the aisles of the show and stood two-deep at booths for assault weapons and ammunition clips.
At all three shows the attendees were overwhelmingly white men, with some women and very few ethnic minorities.
GUN SHOW 'LOOPHOLE'
Thousands of guns shows are held in the United States every year. Under federal law, licensed dealers must conduct a background check before selling to a buyer at a gun show.
But in what critics call a "loophole," which some gun control advocates hope to close, unlicensed collectors and other private sales do not require a background check.
A 2009 undercover investigation at seven gun shows by the city of New York found that 63 percent of sellers failed an "integrity" test by selling a weapon to a buyer who admitted he probably could not pass a background check.
Pennsylvania dealer Donley said on Saturday that since the Connecticut shooting the telephone wait for dealers to get through to the State Police unit that provides background checks has increased from 15 minutes up to 45 minutes.
While most people interviewed at the shows were not in favor of gun controls, not everyone opposed some regulation.
Bruce Abernathy walked away with an assault rifle after sitting through a 30-minute background check at the Texas show.
"There should be more strict background checks," said Abernathy, a Dallas resident. He said there should be a 30-day waiting period to buy weapons and a thorough background check that includes five references. For example, if weight loss is a major goal for employees, a company could support that by providing a meeting room for a weight management program such as Weight Watchers at Work or TOPS (Taking Off Pounds Sensibly), an action that would cost next to nothing.
If employees are looking to improve healthy eating habits, a company can have vending machines stocked with healthy alternatives to the normal fare, such as fruit and pretzels. The cafeteria could also offer a low-fat entree.
To address overall health and disease prevention, an employer can offer health screenings through a local hospital, an insurance provider, or its own medical staff if the company has one. Many people are not aware of potential health problems.
?Only 25 percent of Americans who are of working age know that they have high blood pressure,? said Garry Lindsay, director of Business Partnerships at Partnership for Prevention in Washington, D.C.
Dennis Durr, a senior professional in human resources at the Society for Human Resource Management, suggests tapping into community resources such as the American Lung Association or the American Diabetes Association to promote wellness. Many offer materials or classes addressing specific health concerns that could be offered in the workplace. For example, The American Heart Association offers Heart At Work, an Internet-based program that can be individualized for employees. A subscription costs $300 per year per work site. The Web site includes instruction, a budget planning worksheet, an implementation timeline and posters so that a designated person at the company can coordinate the program.
The benefits of exercise are well-known and irrefutable, and an on-site fitness center is an attractive benefit for employees. It is, however, a costly option, roughly $125 per square foot, and one that involves liability issues, trained staff, and maintenance. Before building a fitness center, a company needs to talk to a qualified professional about liability and workers? compensation. Laws vary from state to state; in some areas, workers who injure themselves in a company fitness center are eligible to receive workers? compensation benefits.
There are less expensive ways to promote fitness that involve little or no liability for employers. A company could institute a lunchtime walking club, with incentives to encourage employees to join. Lindsay suggests that something as simple as providing a changing room and shower so employees who bike to work or exercise at lunchtime would have a place to shower and dress, can encourage exercise.
Offering employees a discount or subsidy for a membership to a local fitness center such as the YMCA removes employer liability and still offers employees a place to exercise. This option could also include family members, an attractive benefit for employees. Businesses can also bring in personal trainers or other fitness professionals to work with employees.
Allowing flex-time hours to fit in exercise is another way to encourage exercise. ?We have people working much longer hours, so we need to make it convenient to them to include an active component in their lives,? said Mier of the American College of Sports Medicine.
The U.S. surgeon general, in concert with health organizations, businesses and other agencies, set forth national health objectives called ?Healthy People 2010.? From those objectives, Partnership for Prevention identified eight work force objectives that it encourages employers to adopt, with the goal of having 75 percent of American companies participating in wellness programs, with 75 percent employee participation, by 2010.
Promoting wellness in the workplace is something every company, regardless of size and budget, can do.
